Process:

Material to be labeled can be provided by the customer or directly ordered from an external vendor by Cisbio Bioassays. Consultation with our staff is provided to develop a labeling approach that addresses the specific needs of the scientist. During the labeling process, our scientists are continuously monitoring sample integrity to ensure quality labeling. Following labeling, the sample is fully characterized to provide the end-user with the appropriate working and quality control information.

Case study
A customer requested the labeling of a protein to be used in a protein-protein binding assay. When the material arrived in our facilities, we set forth to define a labeling strategy that would preserve the binding affinity of the two proteins.

In this instance, we chose to perform a d2-NHS labeling, primarily because the dye is 1kDa and would limit steric hindrance problems. The second reason for choosing this method is that NHS enables rapid and soft labeling. Due to the relatively weak protein-protein binding of the pair, the custom labeling team chose to use high protein concentrations with a high ratio of d2 for this component. This would give the customer a higher working stock with a higher percentage of labeled material to increase the chances of success with the assay. Following a quick protocol optimization, the batch protein was labeled and the remaining unbound d2 was gel-filtrated using Sephadex™ G-25 (GE). Combined with the Europium labeled binding partner, the customer was able to use the d2 labeled material to develop a protein-protein binding assay.

Test Labeling Package:
This option is ideal for researchers who have a valuable sample, want to assess labeling feasibility and efficiency, or are looking to label a small quantity of sample for assay development. Our scientists carry out a small size labeling that generally produces enough material for customers to perform at least 1,000 assay points. This service allows researchers to test the functional performance of a labeled biomolecule in their assay, both at a lower cost and prior to scaling up to batch size labeling. The chemistry and processes optimized during test labeling can be applied to batch labeling protocols during scale-up, thus enabling a streamlined migration from small-scale to large-scale labeling.

Batch Labeling Package:
This service provides custom labeling of large milligram quantities of biomolecules.

Note: Cryptate custom-labeled products are calibrated using a RUBYstar (BMG-Labtech) plate reader. Delivered quantity of the cryptate conjugate is designated as the number of 20µl (final volume) assay points that can be run in a 384-well low volume plate. For XL665, d2, biotin and DNP labelings, delivered quantities of proteins are designated in milligram units and oligonucleotides are supplied based upon OD units.
